Driving Big Data – Integration and Synchronization of Data Sources for Artificial Intelligence Applications with the Example of Truck Driver Work Stress and Strain Analysis

    Get PDF
This paper contributes to the issue of big data analysis and data quality with the specific field of time synchronization. As a highly relevant use case, big data analysis of work stress and strain factors for driving professions is outlined. Drivers experience work stress and strain due to trends like traffic congestion, time pressure or worsening work conditions. Although a large professional group with 2.5 million (US) and 3.5 million (EU) truck drivers, scientific analysis of work stress and strain factors is scarce. Driver shortage is growing into a large-scale economic and societal challenge, especially for small businesses. Empirical investigations require big data approaches with sources like physiological and truck, traffic, weather, planning or accident data. For such challenges, accurate data is required, especially regarding time synchronization. Awareness among researchers and practitioners is key and first solution approaches are provided, connecting to many further Machine Learning and big data applications

Identification of the Tumor Infiltrating Lymphocytes (TILs) Landscape in Pure Squamous Cell Carcinoma of the Bladder

Simple Summary Treatment options in squamous cell carcinoma (SCC) of the bladder are limited and prognosis is poor. In this report we investigated the impact of tumor-infiltrating lymphocytes (TILs) in SCC of the bladder in patients undergoing radical cystectomy. We found that subsets of TILs hold predictive value for OS and PFS. We conclude that TILs might stratify patients with bladder SCC for immunotherapy. Background: Tumor infiltrating lymphocytes (TILs) are known as important prognostic biomarkers and build the fundament for immunotherapy. However, the presence of TILs and its impact on outcome in pure squamous cell carcinoma (SCC) of the bladder remains uncertain. Methods: Out of 1600 patients undergoing radical cystectomy, 61 patients revealed pure bladder SCC in the final histopathological specimen. Retrospectively, immunohistochemical staining was performed on a subset of TILs (CD3+, CD4+, CD8+, CD20+). Endpoints were overall survival (OS), cancer-specific survival (CSS) and progression-free survival (PFS). The Kaplan-Meier method was used to evaluate survival outcomes. Results: Strong infiltration of CD3+ was found in 27 (44%);of CD4+ in 28 (46%);of CD8+ in 26 (43%);and of CD20+ in 27 tumors (44%). Improved OS was observed for strong CD3+ (p < 0.001);CD4+ (p = 0.045);CD8+ (p = 0.001);and CD20+ infiltration (p < 0.001). Increased rates of PFS were observed for CD3+ (p = 0.025) and CD20+ TILs (p = 0.002). In multivariate analyses, strong CD3+ (HR: 0.163, CI: 0.044-0.614) and strong CD8+ TILs (HR: 0.265, CI: 0.081-0.864) were revealed as predictors for OS and the strong infiltration of CD20+ cells (HR: 0.095, CI: 0.019-0.464) for PFS. Conclusions: These first results of TILs in bladder SCC revealed predictive values of CD3+, CD8+ and CD20+

PURPOSE To investigate the expression of the receptor protein ACE-2 alongside the urinary tract, urinary shedding and urinary stability of SARS-CoV-2 RNA. METHODS Immunohistochemical staining was performed on tissue from urological surgery of 10 patients. Further, patients treated for coronavirus disease (COVID-19) at specialized care-units of a university hospital were assessed for detection of SARS-CoV-2 RNA in urinary samples via PCR, disease severity (WHO score), inflammatory response of patients. Finally, the stability of SARS-CoV-2 RNA in urine was analyzed. RESULTS High ACE-2 expression (3/3) was observed in the tubules of the kidney and prostate glands, moderate expression in urothelial cells of the bladder (0-2/3) and no expression in kidney glomeruli, muscularis of the bladder and stroma of the prostate (0/3). SARS-CoV-2 RNA was detected in 5/199 urine samples from 64 patients. Viral RNA was detected in the first urinary sample of sequential samples. Viral RNA load from other specimen as nasopharyngeal swabs (NPS) or endotracheal aspirates revealed higher levels than from urine. Detection of SARS-CoV-2 RNA in urine was not associated with impaired WHO score (median 5, range 3-8 vs median 4, range 1-8, p = 0.314), peak white blood cell count (median 24.1 × 1000/ml, range 5.19-48.1 versus median 11.9 × 1000/ml, range 2.9-60.3, p = 0.307), peak CRP (median 20.7~mg/dl, 4.2-40.2 versus median 11.9~mg/dl, range 0.1-51.9, p = 0.316) or peak IL-6 levels (median: 1442~ng/ml, range 26.7-3918 versus median 140~ng/ml, range 3.0-11,041, p = 0.099). SARS-CoV-2 RNA was stable under different storage conditions and after freeze-thaw cycles. CONCLUSIONS SARS-CoV-2 RNA in the urine of COVID-19 patients occurs infrequently. The viral RNA load and dynamics of SARS-CoV-2 RNA shedding suggest no relevant route of transmission through the urinary tract

An APRI+ALBI Based Multivariable Model as Preoperative Predictor for Posthepatectomy Liver Failure.

OBJECTIVE AND BACKGROUND Clinically significant posthepatectomy liver failure (PHLF B+C) remains the main cause of mortality after major hepatic resection. This study aimed to establish an APRI+ALBI, aspartate aminotransferase to platelet ratio (APRI) combined with albumin-bilirubin grade (ALBI), based multivariable model (MVM) to predict PHLF and compare its performance to indocyanine green clearance (ICG-R15 or ICG-PDR) and albumin-ICG evaluation (ALICE). METHODS 12,056 patients from the National Surgical Quality Improvement Program (NSQIP) database were used to generate a MVM to predict PHLF B+C. The model was determined using stepwise backwards elimination. Performance of the model was tested using receiver operating characteristic curve analysis and validated in an international cohort of 2,525 patients. In 620 patients, the APRI+ALBI MVM, trained in the NSQIP cohort, was compared with MVM's based on other liver function tests (ICG clearance, ALICE) by comparing the areas under the curve (AUC). RESULTS A MVM including APRI+ALBI, age, sex, tumor type and extent of resection was found to predict PHLF B+C with an AUC of 0.77, with comparable performance in the validation cohort (AUC 0.74). In direct comparison with other MVM's based on more expensive and time-consuming liver function tests (ICG clearance, ALICE), the APRI+ALBI MVM demonstrated equal predictive potential for PHLF B+C. A smartphone application for calculation of the APRI+ALBI MVM was designed. CONCLUSION Risk assessment via the APRI+ALBI MVM for PHLF B+C increases preoperative predictive accuracy and represents an universally available and cost-effective risk assessment prior to hepatectomy, facilitated by a freely available smartphone app

A REFERENCE ARCHITECTURE OF HUMAN CYBER PHYSICAL SYSTEMS PART I: CONCEPTUAL STRUCTURE

We propose a reference architecture of safety-critical or industry-critical human cyber-physical systems (CPSs) capable of expressing essential classes of system-level interactions between CPS and humans relevant for the societal acceptance of such systems. To reach this quality gate, the expressivity of the model must go beyond classical viewpoints such as operational, functional, architectural views and views used for safety and security analysis. The model does so by incorporating elements of such systems for mutual introspections in situational awareness, capabilities, and intentions in order to enable a synergetic, trusted relation in the interaction of humans and CPSs, which we see as a prerequisite for their societal acceptance. The reference architecture is represented as a metamodel incorporating conceptual and behavioral semantic aspects. We illustrate the key concepts of the metamodel with examples from smart grids, cooperative autonomous driving, and crisis manage

A REFERENCES ARCHITECTURE FOR HUMAN CYBER PHYSICAL SYSTEMS - PART II: FUNDAMENTAL DESIGN PRINCIPLES FOR HUMAN-CPS INTERACTION

As automation increases qualitatively and quantitatively in safety-critical human cyber-physical systems, it is becoming more and more challenging to increase the probability or ensure that human operators still perceive key artefacts and comprehend their roles in the system. In the companion paper, we proposed an abstract reference architecture capable of expressing all classes of system-level interactions in human cyber-physical systems. Here we demonstrate how this reference architecture supports the analysis of levels of communication between agents and helps to identify the potential for misunderstandings and misconceptions. We then develop a metamodel for safe human machine interaction. Therefore, we ask what type of information exchange must be supported on what level so that humans and systems can cooperate as a team, what is the criticality of exchanged information, what are timing requirements for such interactions, and how can we communicate highly critical information in a limited time frame in spite of the many sources of a distorted perception. We highlight shared stumbling blocks and illustrate shared design principles, which rest on established ontologies specific to particular application classes. In order to overcome the partial opacity of internal states of agents, we anticipate a key role of virtual twins of both human and technical cooperation partners for designing a suitable communicati

Guidelines and Recommendations on Yeast Cell Death Nomenclature

Elucidating the biology of yeast in its full complexity has major implications for science, medicine and industry. One of the most critical processes determining yeast life and physiology is cellular demise. However, the investigation of yeast cell death is a relatively young field, and a widely accepted set of concepts and terms is still missing. Here, we propose unified criteria for the definition of accidental, regulated, and programmed forms of cell death in yeast based on a series of morphological and biochemical criteria. Specifically, we provide consensus guidelines on the differential definition of terms including apoptosis, regulated necrosis, and autophagic cell death, as we refer to additional cell death routines that are relevant for the biology of (at least some species of) yeast. As this area of investigation advances rapidly, changes and extensions to this set of recommendations will be implemented in the years to come. Nonetheless, we strongly encourage the authors, reviewers and editors of scientific articles to adopt these collective standards in order to establish an accurate framework for yeast cell death research and, ultimately, to accelerate the progress of this vibrant field of research
